Leptin is a 167‑amino‑acid protein hormone, approximately 16 kDa in size, encoded by the ob (obese) gene. It plays a central role in regulating body weight, energy balance, metabolism, and reproductive function. Leptin is produced primarily by adipocytes, consistent with the concept that total fat mass serves as a key signal for energy stores. Smaller quantities are also secreted by gastric epithelial cells and the placenta.
Leptin receptors are highly expressed in hypothalamic regions that govern appetite, satiety, thermogenesis, and overall energy expenditure, as well as in T lymphocytes and vascular endothelial cells. Through these receptors, leptin modulates food intake, energy utilization, and adiposity. Beyond metabolic regulation, leptin contributes to hematopoiesis, angiogenesis, wound repair, inflammation, immune function, pubertal maturation, and fetal development.
Research has explored leptin’s involvement in conditions such as obesity, anorexia nervosa, insulin resistance, and hypertension. Leptin also exhibits thermogenic properties and influences enzymes involved in fatty acid oxidation. Notably, severe hereditary obesity in both rodents and humans can result from impaired leptin production.
